Scientific evaluation of Plumbago indica L. for anticancer ingredients, and cytotoxicity actions: Randomized controlled trial.

Plumbago indica L. is a pot plant, and belongs to the plant family Plumbaginaceae Juss.. It enjoys subtropical or warm-temperate climates. The studies were specifically designed to identify anticancer ingredients in methanol-water extract of Plumbago indica L., and evaluate its cytotoxicity actions. For the purpose, methanol-water (40:60 v/v) extract was subjected to conventional preparative high pressure liquid chromatography, and 15 fractions were obtained. Ingredients of obtained fractions were separated, and identified with the help of GC-MS and LC DAD-ESI-MS. Anticancer phenolic compounds such as catechol (C6H6O2), chlorogenic acid (C16H18O9), gallic acid (C7H6O5), 4-methyl catechol (C7H8O2), p-hydroxybenzoic acid (C7H6O3), protocatechuic acid (C7H6O4), pyrogallol (C6H6O3), syringic acid (C9H10O5), syringol (C8H10O3), vanillic acid (C8H8O4), and fatty acids such as linoleic acid (C18H32O2), myristic acid (C14H28O2), palmitic acid (C16H32O2) were separated from different fractions. Fractions were evaluated for their cytotoxicity actions on a rarely studied human hepatocellular carcinoma cell line (HCCLM3). 12 fractions showed good to strong cytotoxicity actions in a range of 200 µg/mL 800 µg/mL, whereas 03 fractions did not show any action even at 800 µg/mL, and no anticancer ingredient was detected from them. 50% growth inhibition values for six most active fractions were calculated, and outcomes were in a range of 85.90 (±3) µg/mL-128.50 (±3) µg/mL. 03 out of these 06 most active fractions were found to contain phenolic content in them whereas all other fractions containing phenolic content did possess cytotoxicity actions that may suggest the importance of phenolic ingredients in anticancer actions. The outcomes also showed a definite dose dependent relationship between amount of fractions, and cytotoxicity actions. It is expected that more studies conducted with the plant can lead to discovery of novel drugs.
