Isolating microsatellites in Lycopodiella (Lycopodiaceae) and assessing microsatellite variation in Great Lakes taxa.

Ongoing systematic studies of the genus Lycopodiella in North America indicate there are several species in the Great Lakes region, including two tetraploids. However the current taxonomy is highly confused and does not reflect all observed morphological variation in the field and herbarium. Recent phenetic analysis of morphological traits has revealed several taxa, some undescribed, as well as several hybrids. Taxa we are investigating include L. inundata, L. margueritiae, L. subappressa, and other undescribed morphospecies, as well as hybrids. We are attempting to use microsatellite variation to evaluate the genetic distinctness of the morphotaxa identified by phenetic analysis, as well as to clarify the evolutionary origins of purported polyploids in Lycopodiella. We used a recently published approach to identify potentially informative microsatellite loci in a "reference" species, Lycopodiella subappressa. We submitted a genomic extract to shotgun genome sequencing and processed the collective reads through QDD software. Considering only dinucleotide repeats of at least 8 bp and tri- and tetranucleotide repeats of at least 5 bp as most likely to express population-level variation, we identified 45 such microsatellite loci, for several of which we designed primer pairs. Trials are currently underway to evaluate successful amplification and confirmation of population- and species-level variation for these microsatellite loci in samples from multiple populations and taxa.
